Promoting Peripheral Axon Regeneration across Nerve Gaps Post Trauma

چکیده

Following peripheral nerve trauma, no technique induces axons to regenerate across a long nerve gaps and restore neurological function. The standard clinical “gold standard” technique for bridging nerve gaps is a graft of pure sensory nerve. However, sensory nerve grafts have many limitations because they only induce good axon regeneration across gaps <2 cm in length, for the nerves repaired <2-3 months post nerve trauma, or for patients <25 y/o. Although many different materials have been tested for their ability in induce axons to regenerate across nerve gaps, none is more effective than sensory nerve grafts. We clinically tested three novel techniques that induce axon regeneration across gaps up to 16 cm in length, for repairs performed >3 years post trauma, and for patients up to 58 years of age. This short review exams some of the techniques tested for their ability to induce axon regeneration and concludes with a brief discussion of the three novel nerve repair techniques.

Permeable guidance channels containing microfilament scaffolds enhance axon growth and maturation.

Successful peripheral nerve regeneration is still limited in artificial conduits, especially for long lesion gaps. In this study, porous poly(L-lactide-co-DL-lactide, 75:25) (PLA) conduits were manufactured with 16 poly(L-lactide) (PLLA) microfilaments aligned inside the lumen. Fourteen and 18 mm lesion gaps were created in a rat sciatic nerve lesion model. To evaluate the combined effect of pe...

Differentially promoted peripheral nerve regeneration by grafted Schwann cells over-expressing different FGF-2 isoforms.

Artificial nerve grafts are needed to reconstruct massive defects in the peripheral nervous system when autologous nerve grafts are not available in sufficient amounts. Nerve grafts containing Schwann cells display a suitable substrate for long-distance regeneration. We present here a comprehensive analysis of the in vivo effects of different isoforms of fibroblast growth factor-2 (FGF-2) on pe...

استفاده از کانال راهنمای عصب در ترمیم عصب محیطی

Although the nerve auto graft still remains the clinical Gold standard in repairing nerve injury gaps, many advances have been achieved to guide regenerating axons across the lesion. Functional recovery after peripheral nerve lesion is depended upon accurate regeneration of axons to their original target tissues. To increase the prospects of axonal regeneration and functional recovery, research...

Paeoniae alba Radix Promotes Peripheral Nerve Regeneration

The present study provides in vitro and in vivo evaluation of Paeoniae alba Radix (PR) on peripheral nerve regeneration. In the in vitro study, we found the PR caused a marked enhancement of the nerve growth factor-mediated neurite outgrowth from PC12 cells as well as their expression of growth associated protein 43 and synapsin I. In the in vivo study, silicone rubber chambers filled with the ...
